Flow Cytometry Steps
- Mixed population of cells and are looking for a particular cell (ex. CD8+ and CD4+ T-cells, but we want CD4+)
- Anti-CD4+ Ab raised in a rabbit are added to cells
- Spun down and washed with saline
- Treated with a secondary Ab with an enzyme tag, created by injecting Ab from rabbit in another animal
- Spun down and washed with saline again
- Cells go through tube 1 at a time, those that floresce with the enzyme tag go one way and the rest of the cells go the other way
